Can I take quests from NPCs?

Quests arrive by themselves as you level up.

Are there rewards for quests?

Yes: experience, currency and items, depending on the quest.

How do I see the details of my current quests?

Ctrl+J.

How do I hide the quest list on the right of the screen?

Open the quest menu (Ctrl+J) and untick Show.

How do I get quests?

Automatically, as your level grows.

How many quests can I have at once?

Up to 5, not counting the Crag Mine war. A sixth quest removes the oldest.

I kill the right mob but the quest does not advance. Is that a bug?

Usually not: you are killing a mob of a different kind. There are ordinary mobs and veteran ones. Check the quest menu.

My friend got a quest and I didn't. Is that a bug?

No. Not all classes get the same quests, and not all classes get quest descriptions.

Do I need quests to get a specialisation?

No. Specialisation comes without quests.
